I wouldn't say it causes damage but it can cause premature wear. The more weight on the unit causes more wear on things like bearings, bushings, ball joints...just about everything.

Internally more weight means more heat generated to move the load...the weak link in the stock motor is the piston/rod/wrist pin...not only the weakest parts...also the least oiling...also the most exposure to the most intense heat...
I am not saying you shouldn't do it...just saying bigger tires and/or more weight is more wear and tear...
